Here is a note from Trey, and I am posting it here on the forum so we can get some additional coverage from the 427 brain trust. This is a symptom we have all either experienced before, or may experience in the future. Therefore, the thread can be of great help to Trey (and someone else too someday).

Hi Paul,
Let me try this one one you! My port engine (which has not given me trouble until the starrboard was fixed!) is acting up occasionaly (3 times now). It will start cold normaly, after runnning for 5 minutes or so, I will shut it off and imediately try to restart, but it won't start and there is some labored starting (starter will come to a complete stop every now and then). Will not restart until next morning. This also happened after idling out to the break water, when I gave it some throttle, it stumbled, died and would not restart (started right up next day). This was my first trip back to the slip on one engine (not fun).
Flooding? I have never known an engine, once running, to flood itself.
Vapor lock? I do not think there was enough time for enough heat to build up to vapor lock (sure acts like it though)
Bad coil, bad pertronix module??? I am going to see if there is condensation under the distr cap next time I go and have time to fiddle......could the spark be grounding out?
Have you ever experienced these symptoms?

Trey,
I'm very glad to hear the good news. Be sure the #7 and 8 plug wires are not touching, as they can cause an induction fire which makes you think the motor is coming out of the engine bay!

Paul,
I am up and running! The marina fixed the carb problem, replaced plugs, wires, and dist cap. Yesterday, I pulled the valve convers and adjusted the valve lash. One exhaust valve was very tight. Absolutely no clearance at TDC on #6. The rest all seemed just a little tight (but had clearance at TDC) and I backed them off just a tad.
Took her out and she ran great. It did start to stumble above 3100 rpm, however when you backed off the throttle, it would come right back. It would run fine at 3000. I think it may be getting starved of fuel. The filter is new, but I am concerned about the amount of sediment I am finding in the filter bowl on filter changes on this tank. I am running 24x22 four blade behind a 2.5:1 paragon (I think).
Thanks for all your help, the valve adjustment was not diffficult at all, and went very well.
